Comparing the reversed-phase HPLC and polar SFC methods to the reversed-phase SFC method, baseline resolution of all three compounds in the mix was obtained in an equal or lesser amount of time using the reversed-phase SFC method. The amount of time required for all three peaks to elute increased in the order of C8 C18 phenyl. Particularly when using the phenyl column, baseline resolution of all three peaks was obtained in half the time (approximately 4 min) required for a simple reversed-phase HPLC method using a C18 column. Another key observation (Figure 3d) was that resolution was dependent upon the amount of methanol in the mobile phase, with resolution increasing as the percentage of methanol decreased. Next, we looked at a more closely eluted pair of compounds (G and H) under the same reversed-phase HPLC gradient conditions explored for the separation of compounds shown previously. A representative chromatogram under the classical C18 reversed-phase HPLC condi- tions (Figure 4) and a chromatogram under SFC conditions (Figure 5) are shown. The trends for the separation of G and H largely reinforced our findings from the separation of A, H, and I detailed previously. The resolution for compounds G and H on the C8 and C18 columns was more equivalent than in the previous SFC example (graphs in Figures 3 and 5), with the phenyl column giving far inferior resolutions.
