Figure 8: Isocratic reversed-phase HPLC run of the six-component mix (20:80 acetonitrile-water with 0.1 % formic acid in each solvent through a 150 mm 4.6 mm Waters Symmetry Shield C18 column at 1.5 mL/min). Elution order = B, A, F, E, D, and C. Figure 7: Isocratic reversed-phase HPLC run of the six-component mix (30:70 methanol-water with 0.1 % formic acid in each solvent through a 150 mm 4.6 mm Waters Symmetry Shield C18 column at 1.5 mL/min). Elution order = B, F, E, A, D, and C. ture (Figure 10). By isocratic reversed-phase HPLC, a reduction in flow rate can enhance resolution by altering the plate number of a column, although not to a great extent (15). In addition, this separation was achieved in far less run time (18 min) than that required using a cyano column under SFC conditions (40 min). Table II shows the elution order of the six-component mixture derived from Figures 8–10. The general indication is that the C18 column under SFC conditions is operation in a normal-phase mode. Conclusion We have demonstrated that, with minimal
