LCGC The Peak - June 2008 - (Page 14) 14 THE PEAK JUNE 2008 Screening and Optimizing High-Speed, HighResolution HPLC Methods in the “Lean Laboratory” Helmut Schulenberg-Schell, Agilent Technologies Laboratory managers are constantly challenged to produce more high pressure liquid chromatography (HPLC) results faster, and of higher quality, without increasing costs. Rapid resolution technologies, including columns that produce high-resolution results at both standard and high operating pressures, are available to help meet this challenge. A careful consideration of the parameters required to generate optimal methods quickly and within regulatory constraints can result in a rational plan for significantly improving the productivity of the HPLC workflow without incurring prohibitive expense. H igh pressure liquid chromatography (HPLC) has for many years been a routine analytical technique built on robust column technology and reliable instrumentation. Each analytical laboratory has accumulated dozens, if not hundreds, of methods typically taking 20–60 min or even longer. However, laboratory managers have precious little time and resources to update their methods to assure that they are keeping pace with improvements in technology. At the same time, they are continually being asked to accomplish more and obtain better results without increasing the size of the laboratory staff. Column technologies and improvements to HPLC system components are available that help realize this goal by enabling laboratories to meet these demands (1). Conscious decisions are required as to how to tune methodologies to best take advantage of these advances in technology using existing
