The subjects and methods Eight healthy, mixed-breed dogs participated in the positivecontrol, blinded, four-period crossover design study. The 21-day study included four treatment periods separated by a seven-day washout period. On Days 0, 7, 14, and 21, each dog was orally administered one of four NSAIDs (firocoxib, carprofen, deracoxib, and meloxicam) at the approved label dosages for osteoarthritis. Ten hours later, each dog was injected with urate crystals in the stifle joint to induce synovitis and lameness. Injections were alternated between the dogs’ left and right stifle. A force plate gait analysis of ground reaction force was performed using a floor-mounted force platform. The device measured in peak vertical force the amount of weight the dog put on the injected leg. Dogs were trotted across the device within 24 hours before each injection to establish a baseline and then 14 and 18 hours (four and eight hours after urate crystal injection, respectively) after each NSAID was administered. During each force plate gait observation, the injected leg was given a lameness score from 0 to 4, which was assessed as follows: • 0 = no lameness • 1 = mild lameness (including toe touch to floor [i.e., weight bearing] on all strides) • 2 = moderate lameness (including toe touch to floor on all strides but was less weight bearing) • 3 = severe lameness (including toe touch to floor on 50% or more of the strides) • 4 = non-weight bearing lameness (including toe touch to floor on less than 50% of the strides) Results Lameness score 4 3 I Firocoxib (5 mg/kg) I Carprofen (4.4 mg/kg) I Deracoxib (1 to 2 mg/kg) I Meloxicam (0.2 mg/kg) 2 1 0 Hour 14 (peak lameness) Hour 18 Time after treatment Figure 1. (No claim of superiority can be made from these data.) At hour 14, when the urate crystal lameness effect was at its peak, dogs administered firocoxib averaged a 0.75 lameness score (Figure 1). Dogs given the other NSAIDs averaged lameness scores of 1.63 to 2.00. The same trend continued in hour 18, with firocoxib averaging 0.25 and the other NSAIDs averaging 0.75 to 1.25.
