Nagy, PhD Induction of fatty liver and inflammation is a hallmark in alcoholic and non-alcoholic steatohepatitis (ASH and NASH). Although hepatocytes are the primary cell type involved in triglcyeride deposition, the critical role of inflammatory modulators in the pathogenesis and progression of ASH and NASH is becoming increasingly apparent. This session will review and help clarify the role of innate immune cells in steatohepatitis, focusing primarily on activity of Kupffer cells, the resident macrophage in the liver, to address the impact of Kupffer cell activation in setting the stage for, and enhancing the progression of, fatty liver and inflammation. Goals and Objectives: n Understand the role of inflammatory and chemotactic mediators produced by Kupffer cells in disease models of fatty liver and inflammation such as ASH and NASH. n Christopher P Day, MD, PhD; . Yee, MD, PhD; Identify intracellular mechanisms and signaling molecules in hepatic macrophages that influence steatohepatitis. Determine the importance of the interactions of the Kupffer cells with other cell types in the liver, both resident cells, such as endothelial cells, stellate cells, and hepatocytes, as well as additional immune cells recruited to the liver in the development of steatohepatitis. Detlef Schuppan, MD, PhD; and Jayant A. Talwalkar, MD The clinical consequences of progressive hepatic fibrosis remain an important cause of morbidity and mortality. Multidisciplinary experts will review the current techniques to measure hepatic fibrosis, as well as the need for novel serum markers and imaging techniques. This expert panel will discuss relevant endpoints, novel targets, and techniques for development of accurate biomarkers and imaging. Developing standard approaches for these concepts is important to progress in drug development to halt or reverse fibrosis progression. Appropriate clinical trial designs will also be discussed and developed.
