filing-now can be accomplished in under two
years," asserts David Bramhill, Ph.D., founder of
Bramhill Biological Consulting. "By incorporating
DNA sequencing into approaches for antibody
discovery, a single campaign can exceed 10,000
hits compared to the paltry 50-100 unique antibodies generated per animal with the traditional
method of mouse hybridomas."
Display systems utilize many recent innovations.
Vast libraries of potentially useful antibodies are
displayed for binding assays employing phage,
DNA, or yeast systems. "Phage display offers advantages in sheer numbers combined with the
robustness and convenience of manipulations,"
suggests Dr. Bramhill. Modern libraries incorporate

antibody repertoire development, and they largely
avoid known "developability" issues. Hence, these
approaches yield antibodies that are far more useful. Many of the best hits are good development
candidates without further engineering.
"Today, progress in DNA sequencing power, microfluidics, detection sensitivities, and assay
throughputs have been combined in various remarkable ways, permitting numerous candidate
antibodies to be rapidly screened," summarizes Dr.
Bramhill. "Simultaneous selection for all of the desired properties of a therapeutic antibody can be
accomplished in a single campaign."

Screening for Monoclonal Antibodies
for Adoptive Immune Cell Therapies
"Immunotherapy, which uses the body's own immune system to fight disease, often uses monoclonal antibodies to target cells of interest," explains Thomas Duensing, Ph.D., chief technology
officer at Intellicyt, a Sartorius company. "Monoclonal antibodies are used extensively in immu-

notherapy. Therapeutic monoclonal antibodies
can augment the body's ability to fight disease."
In general, Dr. Duensing continues, monoclonal
antibodies are used in three different ways to
encourage the body's own immune system to fight
a specific disease such as cancer. First, therapeutic
antibodies can increase the activity of the body's
immune cells by binding to surface molecules in
an approach called checkpoint modulation.
Second, adoptive immune cell therapy co-opts
the body's own immune cells to target and kill
cancer. One form of this adoptive immune cell
