T

cells play a critical role in the adaptive immune response. In naive T cells,
binding of the T-cell receptor (TCR) to peptides complexed with major
histocompatibility complex (MHC) triggers an intricate signaling mechanism
leading to T-cell activation, proliferation, and production of cytokines. Modulating
the TCR signaling pathway with biologics, small molecules or genetic engineering
is highly relevant to many therapeutic areas, including cancer immunotherapy,
adoptive cell therapy, vaccine development, and autoimmune disease interventions. The development of these drugs and therapies mandates the routine use of
assays to profile T-cell function and health.
The need to rapidly monitor immune cell function is addressed by an optimized,
high-throughput multiplex flow cytometry assay using the Intellicyt® iQue
Screener PLUS and the Intellicyt T Cell Activation Cell and Cytokine Profiling Kit.
This assay streamlines the traditional workflow by measuring T-cell phenotype,
T-cell activation markers, cell proliferation, and cell viability while also quantitating
secreted cytokines in a single 10 µl sample in a miniaturized multi-well plate
format. The one-wash assay requires minimal hands-on time.
This type of large-scale high-throughput flow cytometry experiment generates
high-content T-cell activation profiles that provide valuable information for
optimizing immuno-oncology drug development, such as checkpoint inhibitors,
adoptive cell therapy development, other antibody and small molecule screening
applications and manufacturing protocols.
To illustrate the utility and efficacy of the assay in this tutorial, human PBMCs
(peripheral blood mononuclear cells) are treated with three different compounds
