Automating Processes Start to Finish
In a 2018 publication Takeda Pharmaceuticals described a high-throughput flow
cytometry (HTFC) workflow, using the Intellicyt® iQue Screener PLUS and a six-sided
modular HighRes MicroStar robotic system, to take advantage of the screener's
acquisition speed, sample miniaturization, and plate-based data management. The
automated workflow incorporated all of the tedious sample preparation, increased
throughput, and provided precise control for the timing of each step to ensure more
consistent and robust results, plate-to-plate and run-to-run.
The results demonstrated that multiplexing mouse IgG capture beads and cells in
a primary hybridoma screen could potentially add more information during the
decision-making process. The authors indicated that the application of cytokine
capture beads can be used to monitor multiple cytokine levels and cell activation
status simultaneously, and that the HTFC was capable of providing high-content

The system's wide dynamic range allows multiplexed cell and bead assays in the
same well for full cell phenotyping and functional analysis. A suite of available
reagents evaluate phenotypic changes, cell health markers, such as viability and
proliferation, and also quantitate in a bead-based ELISA the amount of secreted
cytokines or IgG. Instead of using multiple assays on different instrumentation
everything can be done in a single well, says Dr. ORourke.
