Applying DOE to Optimize rAAV
Upstream Cell Culture Processes:
An Advanced Approach
Process optimization of recombinant
Adeno-Associated Virus (rAAV) in
suspension cell culture is challenging
because of the complexity and transient
nature of the upstream process. Marie
Enga, a Process Engineer at the French
industrial integrator CPV in INSERM
UMR 1089 at the University of Nantes
explains: " When trying to improve our
rAAV transient production processes in
stirred tank bioreactors, we often have
too many factors to look at. Using Design
of Experiments (DOE), we can extract
information which helps us to make
decisions on which factors to look at first
and then see which of them give us the
highest productivity before we move
onto our next run. " Using this approach
allows researchers to assess the effects
of different factors in parallel, which can
save resources and time.

you can often investigate several factors
at once and evaluate their effect on the

use software to do it there is no bias,
then the results will be objective and are
