How Interpreting and Analyzing Data
From a DOE Can Improve rAAV
Vector Productivity in Sf9 Cell Culture
Spodoptera frugiperda (Sf9) insect cells
and recombinant baculoviruses are
being used at the CPV (French industrial
integrator) at INSERM UMR 1089 in the
University of Nantes to produce recombinant
Adeno-Associated Virus (rAAV) in
suspension culture for in vitro and in vivo
gene transfer applications. To optimize
production yields, researchers at the
University use screening studies followed
by optimization studies to scale-up their
cell culture and production processes in
stirred tank bioreactors.

Proof-of-Concept Study
As a proof-of-concept study, the
researchers at the University of Nantes
wanted to establish optimal working
protocols in a small-scale, stirred tank
bioreactor to produce rAAV vectors consistently
at high titer levels. To achieve a
minimum titer of 10¹² Viral Genomes
(vg)/mL in the cleared lysate of rAAV8
serotype with Sf9 cells, a combination of
small scale reactors (Ambr®

variate data analysis (SIMCA®

were used. With this approach, multiple
parameters can be screened in parallel,
to detect factors that significantly affect
