Additionally, the ratio has a non-linear
behavior on titer indicated by the
Rat*Rat model term. To better understand
in which process window to
operate, a 4D contour plot was used
(Figure 2). The plot columns represent
the media used, while the plot rows
represent the lysis reagent used. Red
indicates a high titer, while blue indicates
a low titer. Figure 2 clearly shows
that the highest titer can be achieved
by using media 2 and lysis reagent 2.
Focusing on that media | lysis reagent
combination (Figure 3), the optimal
region for cell density and ratio can be
defined.
The rectangle in Figure 3 shows a
good operating window to achieve a
rAAV titer of 10¹¹ vg/mL.
