Bollmann explains: " One approach for
process optimization of rAAV is sequential
where many labs simply work step by
step analyzing the effects of one factor
at a time. " However, according to
Bollmann, Design of Experiments (DOE)
studies offer a rational approach for faster
and more accurate process optimization
of rAAV in suspension cell culture. "
Bollmann adds: " DOE allows for greater
efficiency as multiple factors can be
assessed and optimized in parallel, but
since this increases complexity it is easier
to use a specialist software such as
MODDE®
to set up and run a DOE.
The software guides scientists to find the
optimal design based on pre-selected
parameters and helps them rapidly find
the maximum set-point where the interactions
produce the highest titer, as it is
often a combination of factors that produce
the best results. "
Some scientists involved in rAAV manufacturing
find the thought of running a
DOE quite daunting. However, it is possible
to easily set up DOE using the right
tools to reap all the benefits of a DOE
approach. Blouin-Tavel states: " A couple
of years ago we had no experience of
managing a DOE. Now that we work with
DOE software, we realize that it is a great
way to organize our rAAV process optimization.
It
has allowed us to make conclusions
and to find factors that we would not
have identified if we hadn't used a DOE
approach . For example, in one of our optimization
projects with HEK293 cells we
have seen that different plasmid ratios
have had a huge impact on rAAV titer.
This surprised us as it is not something
we expected. Using a DOE means we set
up our experiments without our own biases
in the lab and this is what has led to
these new findings. However, due to cost
constraints at some point we will have to
compromise on increasing plasmid concentration
because it will make our production
too expensive so there will be a
trade-off between cost and titer. "
