Conclusion
Being able to rapidly deliver a consistent, pure, high-titer rAAVbased
product requires scalable, efficient processes across the
production workflow. This article has discussed why these rAAV
production needs require a novel approach that shifts away
from using shake flasks for process optimization and adherent
cell culture for manufacturing. It has detailed that best practice
now includes developing robust suspension cell culture
protocols using DOE software and mini bioreactors for
screening multiple factors in parallel. This approach has been
shown to produce efficient and cost-effective titer increases
of rAAV in suspension cell culture and has the potential to
help deliver safe, efficient, and affordable rAAV-based gene
therapies.
