Inhibition of Spheroid Formation by Virally Mediated RNAi 6
Gene knockdown by RNA interference (RNAi)
has demonstrated tremendous success in rapidly
identifying novel cancer targets. However, most
of this cellular in vitro work has been carried out
on adherent cells grown in 2D, ignoring the role
of the cell-cell or cell-matrix interactions. In Dr.

cancer tumour formation was developed in a

384-well plate to test whether silencing of specific
integrin and receptor tyrosine kinase (RTK) genes
by virally mediated RNAi would inhibit spheroid
formation. This process was quantified by high
content imaging of the 3D spheroid model
post-infection, which showed that lowering the
expression levels of the receptor genes significantly
reduced spheroid formation. These results

confirmed that by combining RNAi technology
with 3D spheroid models and high content imaging,
RNAi libraries can be screened in an HTS compatible
format. This 3D platform may represent a new
screening paradigm from which targets related
specifically to 3D tumour structural growth can
be revealed and exploited for therapeutic
intervention (Figure 2).
