The Challenge of Adapting 3D Cell Cultures
for High Throughput Screening
In recent years a number of different plate and assay formats
have become commercially available to culture cells in 3D, such
as low attachment and hanging drop plates for tumour spheroids
and soft agar systems for cell colonies. To make these assays
compatible with high throughput screening, many of the plates
comply with the SBS standard required for automation and offer
a choice of either 96- or 384-well format. At present, no single
technique has been established as a "gold standard" for drug
discovery and the choice of assay format depends on the needs
of the experimenter (Table 2).
A number of analysis options exist for 3D cell cultures:
spectrophotometric assays can be used to determine parameters
such cell health, viability, and proliferation, but as they generate
whole well intensity measurements they can only provide
single-plex readouts. High content analysis by contrast can
provide additional automated readouts, such as the number,
shape and size of individual spheroids or colonies, as well as
the distribution of different cell types within the 3D structure.
