Forced degradation methods are often
used to rapidly assess biologic stability
and aggregation. Thermal melting (Tm )
and aggregation (Tagg ) measurements are
favorites because they do not require a
lot of sample or time. While thermal
degradation can usually distinguish good
candidates from bad, it is not perfectly
predictive of what will occur at 4 °C or
25 °C. Additionally, it is not unusual for
engineered antibodies and good formulations to have similar or identical Tm or Tagg

measurements, making it difficult to
distinguish the best from the good.
A complementary and under-utilized
measurement is ΔG, the Gibbs free energy.
ΔG quantifies the ratio of folded to unfolded
protein in a given sample at biologically
relevant temperatures.1 The higher the
ΔG, the more stable the biologic. Measuring
ΔG as a function of protein concentration
also provides aggregation propensity and
aggregation pathway information.2
