Aggregation pathway is unique data that indicates whether
aggregation risks come from the native or unfolded state.
ΔG has historically taken a back seat to thermal degradation
measurements because it has required more time and
tedious sample preparation.
Hunky makes ΔG accessible for stability and aggregation
assessment by fully automating up to 96 ΔG measurements in
a single experiment (Figure 1). Automation paired with lower
sample requirements makes ΔG a good fit throughout the
biologic development process. Obtaining quantitative data
with ΔG enables researchers to be more confident in their
biologic stability and aggregation decisions.

Stability and aggregation with ΔG
Biologics exist in equilibrium between the native and denatured
states in solution. This equilibrium is governed by the equilibrium
constant K, where K = e-ΔG/RT (Figure 2). A higher ΔG describes
an equilibrium that favors the native state, and therefore a
protein that is more stable. For example a sample with ΔG = 8.2
kcal/mol would have 1 ppm of denatured protein, while a sample
with ΔG = 4.1 kcal/mol has 1,000 ppm of denatured protein
present (Table). The ΔG for proteins is measured by chemical
denaturation, commonly using urea or guanidine hydrochloride.
